The first player lays down ace, two, joker hearts, The second player adds on three, four, and five of hearts and takes joker. Is this correct? Or must you leave the joker and start a new run?

If the joker represents the 3 (which it seems to in this case), then you can REPLACE the 3 with the joker and take the joker. If you have the card the joker represents, then you replace that joker with the card and take the joker.
