Is everything tightened? I have this issue with my Kenmore when I loosen the wheel to wind bobbins and don't tighten it up right. I loosen it all the way, then re-tighten and am good to go! Hope yours is an easy fix like mine!
If not, there are a bunch of good YouTube troubleshooting videos for Janome. Maybe one of those could help you figure out what is happening.

You could have several issues. One the pressure foot feed is not moving correctly, the wheel on the side of the machine needs to be tightened up or you could have something that is blocking the bobbin and thread from coming up and connecting with the top thread to make your stitches.
